A simple model for presenilins acting as aspartyl proteases with a-secretase activity. The PS
holoprotein (a) is autocatalytically activated by its own aspartyl protease activity (b). The
resulting fragments bind to each other (G. Thinakaran, Chicago, USA) and form the active-
secretase, which cleaves beta amyloid precursor protein (APP) within its transmembrane
domain (c).
